Immunotherapy against hematopoietic malignancies using a patient’s own T-cells modified with chimeric antigen receptor (CAR-T) has exhibited dramatic success. Unfortunately, CAR-T potency against solid tumors has lagged, potentially due to insufficient T-cell expansion and persistence. Further, when attempting to enhance T-cell expansion and persistence, toxicity related to released cytokines and subsequent inflammatory response should be considered. In this webcast, the speaker will describe .. continue reading an approach to improve efficacy in solid tumor treatment using CAR-T cells and address potential toxicity utilizing both activation and safety switches controlled by rapamycin-related drug administration. Assessment by in vivo bioluminescence imaging showed that the activation switch enhanced T-cell proliferation and subsequent anti-tumor efficacy. Additionally, the drug-induced safety switch rapidly eliminated CAR-T cells in a dose dependent fashion.
