Discuss challenges that remain in the discovery, design and optimization of macrocyclic compounds or constrained peptides with good oral .. Read more bioavailability and cell penetration. This conference highlights and questions the promise of synthetic macrocyclic organic compounds as a way to expand therapeutic chemical space while also broadening the types of ‘druggable’ targets. Macrocyclics or constrained peptides have more potential to inhibit flatter surfaces of protein-protein interactions (PPIs) than do smaller molecules which are better suited for ‘traditional drug targets’ such as catalytic sites of enzymes whose ‘pockets or grooves’ fit small molecules nicely. However, it has been a challenge to design macrocyclics with the small molecule advantage of oral bioavailability and ability to reach intracellular targets.
